Replicate 1 and 2 each represents about 100 meristems or flower buds from 5 plants in the field. Replicate 3 and 4 were from a single plant in the greenhouse. Meristems and flower buds were dissected by micro-dissecting forceps under a dissecting microscope. Stand-specific libraries prepared from total RNA and sequences of 51 bp were generated on an Illumina HiSeq2000.
